Foxp3 actively represses the expression of a cAMP-degrading enzyme, phosphodiesterase 3B (Pde3b). Nonetheless, our understanding of the cell-intrinsic mechanistic and molecular links between cAMP levels and Treg cell suppressive function in different tissues remains incomplete. In the present project, we will study the role of Pde3b as well as of the cAMP-mediated transcriptional regulator ICER in Treg cell differentiation and function in tissues such as the intestinal lamina propria and peripheral and central nervous system, as a way to unravel new potential therapeutic targets for autoimmune and inflammatory diseases.
