The central aim of this project is to define the lung tissue-specific regulation of Treg cell development, phenotype and function by eicosanoids using model of type 2-mediated inflammatory diseases. To this end, we will study the distinctive capacities of immunomodulatory eicosanoids to control the differentiation of a stable and functional Treg cell phenotype in vitro (Aim 1). Aim 2 will then investigate the functional characteristics of eicosanoid-modulated Treg cells in vivo in the context of house dust mite (HDM)-induced allergic airway inflammation (HDM-AAI) with a particular focus on prostaglandin E2 (PGE2). In Aim 3, we will mechanistically and functionally elucidate the crosstalk between Treg and PGE2 producing myeloid cells during AAI.
