Due to the clinical need for sustainable treatment of antibody-mediated rejection and the pivotal role of B cells in this context, we will move forward with our well-characterized renal transplantation (RTx) model in rats. We plan to investigate the synergistic effects of a T-cell depleting and anti-BAFF receptor therapy for prevention of rejection in pre-immunized recipients as well as on the secretion of Dickkopf 3 (DKK3), which has been recently characterized regarding its relevance in transplantation medicine. As a perspective, we want to examine the effects of DKK3 silencing in an experimental RTx model with regard to prevention of development of interstitial fibrosis and tubular atrophy.
