Molekulare Charakterisierung und Klonierung der Mutante Knubbelkopf des Medakafischs

Knubbelkopf (knf) is one of 47 mutants in a pilot mutagenesis screen of medaka fish (Yamamoto, T.1975) that is affected in eye development. This mutation will be extensively characterised, using standard techniques established in J. Wittbrodt's lab, mapped on a high resolution genetic map, and then be cloned using positional cloning techniques. Moreover, in an new mutagenesis screen already initiated at the EMBL further mutations affecting eye development will be isolated and their role in the complex network of eye formation will be investigated. Furthermore, a high resolution (more than 600 meiosis) genetic map that is indispensable for positional cloning attempts will be constructed using interspersed repeat sequence (=IRS) PCR technology, and linked to another, high density (more than 600 marker) genetic map of the Medaka fish.
