The proposed work deals with the structure and dynamics of the interphase nucleus and the intranuclear transport of macromolecules in vivo. The spatial distribution and mobility of essential proteins of the cell nucleus, especially transcription factors and chromatin constituents, is going to be studied quantitatively by means of advanced fluorescence techniques such as fluorescence correlation spectroscopy, fluorescence photobleaching and single molecule detection, on a new laser scanning device of our own construction. In collaboration with other participants of this priority program, a detailed three-dimensional picture of the mobility of these proteins shall be developed and correlated with the nuclear morphology. In addition, the interpretation of the experimental data will be supported through computer models of the territorial organization of chromosomes.
