Identification and functional analysis of a novel LDL-receptor-related protein (LRD) in the murine central nervous system

The aim of my research proposal is to identify novel proteins involved in neuronal migration during the development of the central nervous system (CNS) using the cerebellum as a model system. Detailed knowledge about the development of the CNS is a prerequisite for the understanding and treatment of many CNS diseases. As gene function is often conserved between invertebrates and vertebrates, I will use a comparative genomics approach to identify functional homogeneity of C. elegans genes involved in neuronal migration. In particular, a novel member of the LRP (LDL-receptor-related protein) family, MIG-13, is a key determinant in the positioning of anteriorly migrating neurons in C. elegans. Besides their function in cholesterol homeostasis, LRPs play a role in a variety of signaling events especially during the development of the CNS. I plan to analyze the function of the murine homolog of this novel LRP using in vitro migration assays. The long-term goal of the research proposal will be to identify extracellular ligands of the orphan receptor. The functional analysis of this novel receptor and its family members will further elucidate the biology of LRPs and open new perspectives in the analysis of neuronal migration during CNS development.
