Final Report Abstract

Pioneers work was done during the course of this project not only establishing transformation methodology for pearl millet but stably expressing antifungal genes in transgenic pearl millet which confer resistant to major phytopathogens. We have presented novel approaches and results in pearl millet in vitro culture and genetic transformation. Nevertheless, pearl millet research is still at its beginnings, and the difficulties we have faced are yet to be dealt with. Establishing plant in vitro culture and transformation is a laborious and long termed process and is influenced by many factors. Concerning pearl millet, genotype dependency, long generation periods and lack of hints and information in literature complicated the research work. Additionally, difficulties in standardizing growth conditions in Europe (e.g. in green house, which is strongly influenced by external weather conditions, especially the winter period) lead to sub-optimal donor plant material, which impacts the response of in vitro cultures. Although further optimization in transformation methodology needs to be done, the project team were indeed first world wide in generating transgenic and fungal resistant pearl millet.
