Final Report Abstract

Die Aldosteronsynthase (CYP11B2) stellt ein neues pharmakologisches Target zur Behandlung von Herzinsuffizienz und Myokardfibrose dar, das erstmals von uns propagiert wurde. Durch die Förderung der DFG war es uns möglich die ersten hochaktiven und äußerst selektiven Hemmstoffe dieses Enzyms zu entwickeln. Durch rationale Weiterentwicklung dieser Verbindungen gelang es uns kürzlich Substanzen zu erhalten, die an der Ratte in vivo aktiv sind, d.h. den Aldosteronspiegel absenken (proof of concept) sowie an derselben Spezies ein gutes pharmakokinetisches Profil zeigen. Damit sind die ersten Voraussetzungen erfüllt, die an ein Therapeutikum zur Behandlung von Herzinsuffizienz und Myokardfibrose zu stellen sind.
