The new project A25N investigates the role of cargo de-ubiquitination by the ubiquitin hydrolase Doa4 as an on-switch for the internalization of cargo by the ESCRT-III/Vps4 machinery into intraluminal vesicles in the endocytic pathway. Interactions between endocytic cargo, ubiquitin, Doa4 and ESCRT-III/Vps4 will be reconstituted in vitro to determine their effect on ESCRT-III assembly structures and dynamics. Based on these effects, Doa4 will be engineered into a tool allowing to synchronize endosomal membrane remodeling for in situ structural biology.
