DNA damage can interfere with transcription and defects in transcription-coupled repair accelerate the aging process in humans. We have established C. elegans as organismal model for studying the consequences of transcription-blocking DNA damage. Here, we will investigate the role of the SRCAP orthologue ssl-1 in regulating the response mechanisms to transcription impediments. We will assess how the regulation of gene expression and transcription elongation is influenced by SSL-1-dependent deposition of the HTZ-1/H2A.Z histone.
