Alternative polyadenylation (APA) is an RNA processing mechanism that generates transcripts with different 3′-ends for the same gene. Recent studies have shown that APA contributes to cellular senescence through its influence on gene expression. We will investigate whether APA undergoes upstream (i.e., changes in splicing or transcription) or intrinsic (i.e. changes in the APA machinery) loss of fidelity associated with cellular senescence by combining poly(A) sequencing with eCLIP of key regulatory proteins and mapping of their protein-protein interactions to identify generalizable points of intervention.
