B03 focuses on calcium signalling, specifically the TPC1 cation channel in the vacuole. The project aims to understand the proteoform-specific regulation by cytosolic calcium and phosphorylation, emphasising selectivity. Using imaging techniques, the impact of TPC1 proteoforms on Ca2+ signal generation, decoding, and information propagation to plant parts will be analysed. Using electrophysiology, phosphoproteomics and imaging, the project aims to reveal insights into how systemic regulatory components' natural variation aids adaptation to environmental stress.
