Einfluß genetischer Variation in beta-2-adrenergen Rezeptoren auf die Regulation des Blutdrucks im Menschen

Common polymorphisms in the gene encoding the (2-adrenergic receptor have been associated with interindividual differences in blood pressure and the diagnosis of essential hypertension. Preliminary human and in vitro studies have demonstrated that these polymorphisms alter vasodilator responses to (2-agonists, which may modulate the pressor response to endogenous catecholamines during sympathoexcitatory stimuli such as mental stress. The magnitude of the pressor response to sympathoexcitation is a risk factor for the development of hypertension. The aims of this project are: (1) to genotype healthy individuals for genetic variation in the (2-receptor; (2) to study the influence of genetic variation on the systemic effects of a (2-agonist before and during ganglionic blockade in order to dissect vascular responses due to (2-receptor stimulation from those due to baroreflex activity; and (3) to explore how genetic variation in the (2-receptor influences endothelium-dependent and -independent vasodilator responses and pressor responses to sympathoexcitation. These investigations are expected to advance understanding of how genetic variation in the (2-adrenergic receptor influences vascular control and blood pressure responses in humans and contributes to intermediate physiological patterns relevant to the pathogenesis of hypertension.
