Mitochondria contain a unique translation machinery, which consists of the mitochondrial ribosome (mitoribosome) and specialized translation factors. Defects in mitochondrial translation such as mitoribosome stalling lead to a decline in cellular energy production associated with severe human diseases. To resolve stalled mitoribosome complexes, specific rescue systems are required, which are still ill defined in human mitochondria. Here, we aim to decipher the molecular machineries and mechanisms of rescuing mitoribosomes stalled under distinct conditions, and to dissect the interplay between the mitoribosome and the quality control machinery that prevents proteotoxic stress.
