Antibodies, cytokines and over more than 1000 different proteins interact with and shape biological hydrogels. For only few proteins the interaction with mucins or the glycocalyx has been explored. This project aims to investigate hydrogel interactions of natural proteins (e.g., antibodies, cytokines) as well as artificial fluorescent model proteins and nanobodies using a battery of biophysical techniques. We expect to identify molecular features which decide on protein retention or diffusion in hydrogels. From these outcomes combined with protein-polymer engineering strategies, this project will also address higher or lower mucus transport of proteins depending on potential needs for drug delivery strategies.
