The microscopy system combines Total Internal Reflection Fluorescence (TIRF) and spinning disk (SD) microscopy technologies. This advanced setup aims to enhance the imaging of biological samples in both 2D and 3D formats. Key goals of the system include: 1) High Resolution and Sensitivity: Utilizing TIRF, the system provides high-resolution images with excellent signal-to-noise ratios by focusing illumination at the interface between cells and glass-like materials. This technique minimizes photo-bleaching and cellular damage, crucial for studying delicate biological samples. 2) Rapid and Controlled 3D Imaging: The SD component allows for fast, controlled 3D imaging, overcoming the slower, point-by-point image acquisition of traditional confocal microscopy. This is achieved through a mechanism of rotating disks with multiple pinholes, enhancing light throughput and reducing phototoxicity during prolonged imaging sessions. 3) Dual-Channel Live Imaging: By integrating both TIRF and SD technologies on the same platform, the system facilitates quick switching between different imaging modes. This is particularly valuable for live imaging experiments, enabling detailed observation of plasma membrane dynamics and cellular processes in real time. 4) Environmental Stability: The inclusion of an environmental chamber ensures that experiments can be conducted under controlled temperature conditions, vital for consistent and reproducible results. 5) Future Expandability: Plans are in place to further enhance the system's capabilities by adding a super-resolution module. This will allow for even more detailed imaging of cell interfaces and intracellular structures. This microscopy setup addresses specific technical needs such as dual cameras and a wide-range piezo. It is designed to significantly advance research by providing a versatile, high-performance imaging tool for complex biological investigations.
