This project investigates brain tissue replacement therapy using human stem-cell-derived cortical organoids implanted into the infarct core after stroke. By combining human cortical organoid transplantation with advanced two-photon calcium imaging in behaving mice, the project assesses how grafted tissue integrates into vascular and neural networks of the peri-infarct area. It further explores the role of graft composition and aging on recovery outcomes, aiming to develop a foundation for personalized regenerative strategies for chronic stroke patients.
