This project will characterize the underlying mechanism between IOP, impaired iron homeostasis, microglial activation, and neuroinflammatory signaling related to a vicious circle of retinal ganglion cell loss. The investigation of IOP elevation and cellular neuroinflammation will be performed in vitro in retinal tissue as well as in in vivo models (i.e. episcleral vein cauterization). Analysis of aqueous humor, retina, and serum from glaucoma patients will follow. Murine models of hepcidin deficiency and hepcidin increase will be exposed to the IOP models, thereby revealing signaling mechanisms and offering therapeutical opportunities.
