Co-translational processing in eukaryotes ensures proper folding and targeting of nascent proteins by ribosome-associated factors. NAC, positioned at the ribosomal exit tunnel, recruits biogenesis factors to nascent chains for efficient processing. Under translational stress, ribosome stalling leads to accumulation of defective proteins. This triggers the activation of RQC pathways that target stalled chains for degradation and promote ribosome recycling. Recent data show that RNF25 ubiquitylates NAC under heat stress, suggesting stress-responsive regulation. This modification may coordinate NAC function with RQC activity at the ribosome. The project aims to dissect NAC-RQC crosstalk and define RNF25’s mechanistic role in this process.
