Ribosome biogenesis is a fundamental and very complex process in all living cells. Formation of the two eukaryotic ribosomal subunits requires all three nuclear transcription machineries, four ribosomal RNAs, more than 70 ribosomal proteins, more than 150 non-ribosomal factors and about 100 non-coding small RNAs. The huge efficiency of ribosome biogenesis - a growing yeast cell produces more than 40 ribosomes per second - demands an optimal coordination of all participating processes and a major amount of cellular energy. Therefore, regulation of ribosome biosynthesis is tightly regulated.
The long-term goal of the Research Unit is to determine the structural basis of the machineries involved and to dissolve and to connect single steps of ribosome biosynthesis. Thus, it should be possible to gain a comprehensive view, how the complex processes leading to mature ribosomes are regulated and coupled. Our Research Unit opens the particular perspective to study the coupling of chromatin structure, transcription, maturation and transport of ribonucleoprotein complexes (RNP) using the biogenesis of ribosomes.
