Role of platelets in liver injury and regeneration (A16)

In this project three aspects should be investigated in experimental mouse models to characterize the role of bile acids for platelet-mediated effects on cellular responses upon liver injury and regeneration. In a first step, we want to understand the role of the bile acid receptor TGR5 in haemostasis and thrombosis. Secondly, we plan to characterize bile acid-induced TGR5 activation at the platelet membrane and the mechanisms and consequences for platelet activation defects after liver injury. Thirdly, we want to determine the impact of platelet TGR5 for platelet-mediated inflammation and cell survival after liver injury. In a first translational approach, we plan to identify the coincidence of liver injury and alterations in platelet signalling in patients with cholestatic liver disease.
