Critical transitions in the eukaryotic cell cycle are entry into S-phase and the onset of mitosis. Cdc25 phosphatases play important roles in regulating these transitions by activating cyclin-dependent kinases through dephosphorylation of inhibiting threonine and tyrosine residues. In human cells, three Cdc25 phosphatase family members have been described, Cdc25A, Cdc25B and Cdc25C. The aim of this study is to carry out a detailed analysis of the molecular mechanisms underlying the regulation of the G1/S transition by Cdc25A in human cells. Isolation and characterization of proteins involved in the regulation of Cdc25A will contribute to better understand the mechanisms of S-phase onset. Furthermore, a study on the function and regulation of human Cdc25B phosphatase during S and G2-phases will be performed. In addition, a novel putative member of the Cdc25 phosphatase family will be functionally characterized.
