Free radicals in mechanical trauma to the muscle. Identification of the contributing species, mechanisms of their formation, and their effects on metabolic and immunologic functions

Reactive oxygen species, nitrogen monoxide (NO) and reactive nitrogen species derived from NO are decisively involved in a great variety of cell and tissue injurious processes including mechanical injury to the brain and other tissues. Here we will study the participation of these free radicals in the mechanical trauma to the muscle. Studies will range from cell culture experiments via mechanical injury to the rat hind limb to analysis of probes from patients with soft-tissue trauma. A great variety of analytical methods will be used including different in vivo microscopic approaches. After analyzing the time sequence of the formation of the different free radicals, we will unravel the underlying pathogenetic mechanisms of their formation and delineate their effects on selected metabolic and immunologic functions in close cooperation with other applicants of the priority program. In a later phase of the project, we are planning to initiate a therapeutic study based on the experimental data.
