SFB 469:  Induction and Inhibition of Proteolysis-Mediated Processes in Inflammation and Neoplasia

A complex dysregulation of proteases and their inhibitors seems to play a central role particularly in diverse pathologic reactions of the organism. Therefore, the collaborative research centre 469 focuses in two closely related research sectors to contribute to the further evaluation of proteolysis-induced pathomechanisms in inflammatory and malignant diseases, and their possible therapeutic treatment.Projects in the basic research sector A focus on the determination of three-dimensional structures of proteinases and their specific inhibitors, the subsequent structure-based design of novel inhibitor molecules with specifically introduced desired characteristics, and the functional characterization of these inhibitors. These studies are expected to extend the knowledge to a point suitable for an efficient inhibitor design, and to result in the production of compounds useful as tools for the analysis of pathomechanisms and as lead compounds for the development of drugs effective in clinical trials.Projects in the clinically oriented sector B are investigating whether the alterations of protease/inhibitor systems in inflammatory and neoplastic diseases are similar or distinct. In particular, studies are focused on the proteolysis associated with the penetration of inflammatory and tumor cells through basement membranes and extracellular matrix, and on the application and evaluation of novel inhibitory strategies (e. g. direct application of inhibitors, inhibitor overexpression in transfected cells, antisense-nucleotides to protease downregualtion) in cell culture and animal models.
