The role of chronic hypoxia in affecting biological systems is rarely known, although affecting expression of genes via tristetraprolin. We determine the post-transcriptional regulation of genes via tristetraprolin using the individual-nucleotide resolution UV cross-linking and immunoprecipitation- and NGS-method, to understand the role of affected genes in macrophages. In addition, HIF-1 impairs the assembly of complex I of the mitochondrial respiratory chain by inducing β-TrCP (β-transducin repeat-containing protein) and degrading the assembly factor TMEM126B. The relevance of this observation towards the functional role of mitochondria in determining macrophages and tumor cells will be evaluated in vitro and in vivo.
