Coxsackievirus B3-induced modulation of K-channels in viral myocarditis associated arrhythmias

Goal of the proposed research project is the characterisation of pathogenetical mechanisms of potentially lethal CVB3 induced arrhythmias in human iPSC derived cardiomyocytes and human myocard. The CVB3 induced functional changes of cardiac ion channels and associated basic molecular mechanisms in human cells shall be studied in HEK cells, novel iPSC-cardiomyocyte lines and infected human heart in vitro and ex vivo. Specific bullet points of the project are:1. Verification of CVB3 mediated ion channel modulation in HEK cells, iPSC derived cardiomyocytes and infected human tissue. 2. Proteomic analyses of CVB3-transgenic mice in comparison to control mice by mass spectrometry.3. Analyses of molecular details of ion channel modulation by CVB3 in HEK cells, iPSC derived cardiomyocytes, infected human heart and CVB3 transgenic mice.4. Evaluation of identified proteins as pharmacological targets in iPSC derived cardiomyocytes.5. Integration of identified effects in mathematical action potential simulations.
