Generation of breeders for setting up mutant mouse colonies by priority programme members

This Central Service Project generates and maintains mutant mice that are relevant for mast cell research. Breeder pairs and limited numbers of offspring from several mutant mouse colonies as well as advice and guidance on how to work with these mice is and will be provided to interested participants of the priority programme. Specifically, natural mouse mutants that are deficient for mast cells (MCs) will be bred and kept at the mouse facilities of the Department of Dermatology and Allergy at Charité – Universitätsmedizin Berlin. These mutants include WBB6F1-KitW/W-v (W/Wv) mice, C57BL/6-KitW-sh/W-sh (sash) mice and WCB6F1-KitSl/Sl-d (Sl/Sld) mice. In parallel, the mouse facilities of the Institute of Immunology at TU-Dresden will generate and keep colonies of several lines of Cre/loxP-based mouse mutants including mast cell-specific Cre-expressing lines, mice with MC-specific inactivation of various genes, novel models of inducible or constitutive MC deficiency and MC reporter strains in which MCs express fluorescent proteins. These mouse mutants are essential for the identification and characterisation of novel MC functions. The majority of projects within the priority program will, at some point, require the use of such mutant mice. As these mice are not readily available from commercial or other sources, this service project will greatly facilitate the work of most members of this priority program. In addition, this central service project will significantly cut the time and costs involved in obtaining these mice for the projects of the program. Finally, the expertise available at the two centres that contribute to this core facility will have all projects working with mutant mice benefit from this central service projects, as the applicants and their teams provide advice and protocols for designing and running in vivo using MC mouse models.
