Molecular analysis of intestinal stem cell associated genes in colorectal cancer (CRC)

Zusammenfassung der Projektergebnisse

Extensive functional analyses of the 3 candidate genes for colorectal tumor progression, IGF2, CITED4 and KLF6, confirmed their role in cellular processes that are associated with tumor development and dissemination. The most intriguing results were achieved by knockdown (IGF2, CITED4) or knockout (KLF6) of the respective candidate genes. The data confirmed the concept of a role in regulation of cell proliferation (IGF2, CITED4), cell adhesion (IGF2, KLF6) and migration (KLF6). Microarray based expression profiling subsequent to genetic manipulation of each of these candidate genes highlighted a further role of these genes in a number of unexpected cellular pathways and processes including cell/cell contacts (IGF2, CITED4) and the cytoskeleton (CITED4). Altogether, the expression profiles of the respective cell lines revealed a number of deregulated pathways that warrant further analysis in colorectal tumor models.
