Molecular mechanisms of hepatitis B virus persistence in chronically infected human chimeric mice (A05)

Functional cure of hepatitis B virus (HBV) chronic infection will require the identification of factors affecting stability and activity of the virus persistence form, the cccDNA, as well as enhancement of host immune responses. Using HBV infected human liver chimeric mice we will study the virological and cellular consequences of human hepatocyte proliferation. Moreover, to understand virus-host interplay leading to HBV persistence and pathogenesis we will investigate the role of host factors and cytokines in cccDNA regulation, as well as the consequences of HBV infection on bile acid metabolism. Furthermore, we will perform adoptive transfer of human immune cells to investigate non-cytolytic antiviral effects, as well as whether HBV alters the expression profile and function of immune cells.
