Molecular characterization of the biocontrol activity of Pseudozyma flocculosa

The basidiomycetous fungus Pseudozyma flocculosa is a natural inhabitant of the phyllosphere and has been isolated as potent biocontrol agent against fungal pathogens of plants. The active principle of this biocontrol activity is hypothesized to be mediated by the release of an antifungal glycolipid, flocculosin. However, the molecular and genetic basis of glycolipid production and secretion is largely unknown in P. flocculosa. In the related fungus Ustilago maydis, we have recently characterized a biosynthetic gene cluster, which contains all genes required for the efficient production and secretion of ustilagic acid, a highly similar glycolipid. In this proposal I want to elucidate the genetic and molecular basis of biocontrol activity in P. flocculosa. For this aim, the presumed flocculosin biosynthesis gene cluster will be isolated and molecularly characterized. The biosynthesis pathway will be analysed by mutation of single open reading frames and chemical characterization of secreted intermediates. This approach of metabolic engineering will not only allow the understanding of the structural basis of the antagonistic activity but will also help to improve the biocontrol potential of P. flocculosa.
