A04 defined that the gastric pathogen H. pylori induces classic NF-κB activation via IKK-TAK1/TABs/TRAF6 leading to an inflammatory response. Screens using E3 ubiquitin ligase siRNA libraries identified F-box/SOCS-box E3 enzymes ASB3 and TRIM28, which contribute to NF-κB activation. Further, RING-finger/RING-finger-like ubiquitin-editing enzyme A20 suppressed NF-κB activity and apoptotic cell death. During the 3rd funding period, A04 aims to elucidate the entire type 4 secretion system-directed membrane-proximal NF-κB signaling using biochemical and microscopic techniques.
