In the previous funding period we have used a selective ablation model to show the appropriate integration of neurons transplanted from fetal cerebral cortex into the adult murine visual cortex. In the present project we aim first to move this approach towards more clinically relevant models such as traumatic brain injury also in older mice and examine myelination of the transplanted neurons, a key question as regenerating mature neurons fail to be myelinated. In the second project part we will examine the integration and axonal myelination of new neurons obtained by direct reprogramming from local reactive glial cells, based on the breakthrough in this approach after traumatic brain injury we achieved in the last funding period using the read-outs from in coll. with A08.
