Identification of the (1-6)-ß-glucan receptor

Recent evidence suggests that (1→6)-β-glucans are recognized by an unidentified innate immune receptor and are more potent immunomodulators in humans than (1→3)-β-glucans. Previous research has focused on (1→3)-β - glucan. The discovery of Dectin-1, the (1→3)-β-glucan receptor, had a major impact on our understanding of the recognition and response of mammals to fungal pathogens. However, little is known about the immune recognition of (1→6)- β-glucans. We will identify the (1→6)-β-glucan receptor, using a retroviral based cDNA library, previously created by Dr. Gordon Brown to identify the (1→3)- β-glucan receptor Dectin-1. This retroviral library will be integrated within the genome of recipient cell lines and the cells will be screened for the receptor using labeled (1→6)-β-glucan. The receptor gene sequence will be amplified by PCR. The resulting DNA will be sequenced. The amplified DNA will be ligated into a cloning vector and transformed into an eukaryotic expression vector. A recipient tissue cell line will be used to express the protein of interest and to confirm the proteins receptor function. Saturability and dose dependence as well as specificity of binding will be established. This project will have important impact on understanding the role of (1→6)-β-glucan in human response to fungal pathogens.
