The purpose of the project is the development of a MSI-platform for high resolution analysis of spatial localization and dynamics of the structurally and functionally most variable messenger molecules in the CNS, namely neuropeptides. While the new field of neuropeptidomics allows the neuropeptides from a sample to be characterized, their spatial dynamics are poorly understood because of a lack of measurement tools. There is a critical need for improved mass-spectrometrybased imaging/localization. The maintenance of a sufficient resolution in combination with high sensitivity as typical of conventional direct tissue profiling is a challenge in MSI of neuropeptides. The applicants cover longstanding experience in Biochemistry, Neuroscience, and Genetics. M most importantly in the context of the proposal, they have a strong background in MALDI-TOF mass spectrometry and peptidomics, and focused on the development of methods for the analysis of neuropeptides in identified and dissected neurons, in Drosophila and other insect nervous and peripheral tissue or method development in MSI itself (Sweedler). The proposal is divided in two subprojects: 1) Development of MSI for the analysis of immediate compensation mechanisms within the mammalian peptidergic system. 2) Development of MSI-approaches for the functional analysis of neuropeptide dynamics in development and behaviour of a social insect.
