Regulation of the activity and localization of the motorprotein Kif18A by posttranslational modifications (B07)

The overarching aim of this project is to better understand how posttranslational modifications and selective degradation by the ubiquitin-proteasome system adjust protein function to the needs of specific cell cycle stages. For these studies we will use the mitotic kinesin Kif18A as a model substrate. Kif18A is essential for the correct segregation of chromosomes, undergoes multiple posttranslational modifications and is degraded as cells exit mitosis. Degradation of Kif18A starts at anaphase onset but substantial levels of Kif18A remains until the G1 phase. Thus, one focus will be on the mechanisms underlying the complex degradation pattern of Kif18A. Studies from different organisms indicate that Kif18A regulates microtubule dynamics as cells exit mitosis. Therefore, the second focus of this project will be to identify a potential post-metaphase function of human Kif18A. These studies will be key to better understand the mechanisms regulating the partial, cell-cycle specific degradation of Kif18A and identify a potential post-metaphase function of Kif18A.
