The working hypothesis of this project is that after partial hepatectomy (PHx) an enhanced blood flow stretches the hepatic endothelial cells and thereby releases endothelial cell-derived growth factors to initiate liver growth. Conversely, an enhanced shear stress acting on the endothelium results in release of growth-inhibiting factors that stop liver growth and can lead to liver damage. Here, mechanically regulated genes and secreted proteins will be identified in hepatic endothelial cells, mechanistically analysed, and functionally characterized in vitro and in vivo.
