Nucleic acid nanoagents utilizing the intracellular microRNA-200c machinery for switching functions in breast cancer cells (B04)

MicroRNA 200c down-regulates genes that play a role in aggressive forms of breast cancer. Recently established tools, such as breast cancer cells with either genetic miR-200c knock-out or inducible miR-200c, tumor-targeted synthetic 200c nanoagents, and novel miR-200c sensors based on fluorescent light-up aptamers will be applied for time-resolved analysis of effects of miR-200c modulation on epithelial to mesenchymal transition, 1D- and 2D-cell migration, and chemoresistence. The expected therapeutic miR-200c effect of reducing tumor growth and metastases will be tested in 3D-tumor cell spheroids and in vivo tumor models.
