EXC 114: Zentrum für Integrierte Proteinforschung (CIPSM)
Zusammenfassung der Projektergebnisse
Proteine sind Träger von Funktionen innerhalb lebender Zellen und daher von zentraler Bedeutung für das Leben. Zu Beginn des Clusters wurde deutlich, dass Proteine auch die Pharmazeutika der nächsten Generation sein werden und in der Tat sind Proteine heute in der Krebstherapie nicht mehr wegzudenken. Das Ziel des Exzellenzclusters CIPSM war es diese Entwicklung zu begleiten. Zu diesem Zweck etablierte der CIPSM Cluster sechs Forschungsfelder. Im ersten Feld, Protein-Biophysik, untersuchte CIPSM die Kräfte, die es Proteinen erlauben, sich zu falten oder zu entfalten. Im zweiten Forschungsgebiet wurden die Prinzipien der Proteinfaltung und Missfaltung studiert. Das dritte Forschungsfeld war der Untersuchung der dreidimensionalen Struktur von Proteinen gewidmet mit Hilfe der Röntgenstrukturanalytik, der Kryo-Elektronenmikroskopie und der NMR- Spektroskopie. Im vierten Forschungsfeld bündelte CIPSM das Themengebiet Protein-Nukleinsäure-Interaktionen, um die Regulation der Transkription zu untersuchen. Im fünften Forschungsgebiet wurden neue chemisch-biologische Verfahren und Protein-Engineering entwickelt, um Proteine gezielt zu manipulieren. Im Forschungsfeld sechs hat CIPSM Proteine in Krankheitszuständen untersucht mit einem Schwerpunkt auf der Entwicklung der Photopharmakologie bzw. der Untersuchung von Proteinfunktionen in Systemen. Das Thema Systembiologie wurde in München durch CIPSM aufgebaut. In allen Forschungsfeldern gelang es CIPSM wissenschaftliche Durchbrüche zu erzielen, was belegt wird durch über 2400 Publicationen darunter 25 Veröffentlichungen in Nature, 27 Veröffentlichungen in Science und 15 Publikationen in Cell. CIPSM hat darüber hinaus Grundlagenforschung in die Anwendung überführt. Die Aufklärung der dreidimensionalen Struktur der Seidenproteine und die Entwicklung effizienter Expressionssysteme führte zu exzellenten Veröffentlichungen und erlaubte die Gründung der Firma AMSilk GmbH. Neue clickchemische Methoden zur Modifikation von Biomolekülen lieferten hoch zitierte Veröffentlichungen und brachte die Gründung der Firma Baseclick GmbH, die derzeit neue Technologien für das next generation sequencing entwickelt. Das Interesse von CIPSM, hochpotente Antikörper zu entwickeln, führte zur Entwicklung der Kamel-Antikörper und zu den Spin-offs ChromoTek GmbH und Tubulis GmbH. Neue Ideen um die zelluläre Lebensdauer von Proteinen zu beeinflussen, waren die Basis für die Entdeckung des Pasylierungskonzeptes mit der Gründung der Firma XL-protein GmbH. Die Idee fundamental wichtige Forschungsergebnisse in die Anwendung zu überführen, erlaubte es CIPSM eine strategische Allianz mit der Firma Bayer AG einzugehen. Prof. Plischke, ein früheres Vorstandsmitglied und heute Mitglied des Aufsichtsrats gibt im CIPSM Cluster Kurse über Innovationsmanagement, die von ca. 100 Doktoranden besucht werden. CIPSM hat erfolgreich vier Anträge auf neue Forschungsgebäude bewilligt bekommen (Bayerisches NMR Zentrum, BioSysM, CPA und ICEM). Die Gebäude werden die Laborflächen auf den Campus Großhadern-Martinsried und Garching signifikant erweitern, mit einem Investment von über 160 Mio. Euro.
