Genetically modified mice are crucial model systems for B-cell immunology. The Central Service Project C03 has provided transgenic / gene knockout technology for the whole TRR130, which was used for many projects. A new reporter mouse strain was developed, named B-VARIA mice, which allows tracking of B cell populations in vivo. In the next funding period, C03 will establish CRISPR/Cas9 for the generation of mice with defined mutations in several genes of the B cell lineage. The methodology for the application of CRISPR/Cas9 directly in zygotes will be established to induce multiple mutations in closely linked genes in a one-step reaction, to insert by homology dependent repair short loxP DNA sequences for conditional depletion of genes during certain phases of the B cell development and to use longer DNA homology fragments for the generation of complex knock-in alleles like reporter genes, immunoglobulin knock-in alleles, and other conditional alleles.
