Project A9 aims at site-specific, sensitive and quantitative monitoring of protein recognition by supramolecular ligands using Raman spectroscopy. The SERS assay in SP1 realizes the idea of “controlled enhancement of multivalency by nanopositioning”: supramolecular ligands are positioned at defined distances and heights on the surface of a gold nanorod using a DNA origami platform in order to achieve significantly stronger binding to proteins. In SP2 we will establish UVRR spectroscopy as a generic tool for label-free, site-specific and quantitative monitoring of protein recognition by supramolecular ligands using a time-resolved approach.
