Traditional Chemical Biology strategies focus on targeted chemical inactivation of enzymes to elucidate their function within complex biological systems. Recent evidence revealed that many naturally deregulated states leading to severe diseases are caused by conformational defects suggesting an opportunity to develop strategies for the repair of such pathogenic protein conformations. We will use HtrA proteases as models to explore both the traditional and novel concepts to establish innovative approaches for chemical and biological protein inactivation and repair.
