Analysen zur Induktion Allergen-spezifischer Toleranzreaktionen bei Patienten mit Typ-I-Allergien

Zusammenfassung der Projektergebnisse

Aufbauend auf unseren Vorarbeiten war das Hauptziel dieser Studie, eine neuartige therapeutische Strategie zu entwickeln, die mittels einer Allergen-spezifischen Induktion von iTreg durch tolerogene IL-10 DC in der Lage ist, sowohl T-Zellantworten gegen Pollenallergene (z.B. Birke) als auch gegen kreuzreaktive Nahrungsmittelallergene (z.B. Haselnuss) in Patienten mit Birkenpollen- und assoziierter Haselnuss-Allergie zu unterdrücken. Wir konnten sowohl durch in vitro Arbeiten als auch in vivo Studien in einem humanisierten Mausmodell der Typ-I Allergie darlegen, dass durch humane IL-10 DC induzierte Betv1-spezifisch stimulierte iTreg sowohl eine Allergen-spezifische (Birke) als auch kreuzreaktive (Haselnuss) Immunantwort in Form einer sogenannten "Kreuztoleranz" unterdrücken können. Außerdem wurde das Protokoll der Generierung humaner tolerogener IL-10DC optimiert, um eine hoch effiziente Toleranzinduktion zu erzielen.
