A novel algorithmic approach, CaCLEAN, provides functional 3D maps of excitation-contraction coupling couplons in beating cardiomyocytes and thus further enlightens remodeling processes in cardiac diseases. In the heart, specific in vivo activation of Gq-coupled signaling via design receptors (DREADD) alters impulse-generation and -propagation and coordinated delivery. Thus, Gq-coupled signaling directly modifies cardiac electrophysiology such as intercellular communication. DREADDs will advance our understanding of the dynamic modulatory impact of Gq signaling in the in vivo heart.
