SNARE complex function in Ca 2+ -triggered exocytosis (B16)

Project Description

The SNARE proteins syntaxin 1, SNAP-25 and synaptobrevin 2 form the basal fusion machineryduring Ca -dependent exocytosis. According to a popular hypothesis, assembly of the ternarySNARE complex between apposed membranes leads to membrane fusion. We will investigatethe assembly pathway of the complex, by introducing destabilizing mutations into synaptobrevin2. In addition, we will use chimeric constructs between different SNAREs to search forspecializations that allow membrane fusion to be fast. All mutations/chimera will be investigated in a null background using fast electrophysiological techniques.
