The aim of the project is to study the role and regulation of changes in DNA modifications and gene expression, using long-term in vitro cultured patient derived xenograft acute myeloid leukemia cells with and without selective pressure. We plan to look at genome-wide changes and at the conservation, variation and variance of patterns as a function of specific mutations in epigenetic modifiers (DNMT3A, TET2 and IDH1/2) and oncometabolite levels (2HG). The overall aim is to identify and counter epigenetic mechanisms that affect the response or resistance to therapy.
