Zusammenfassung der Projektergebnisse

The aim of the project was to elucidate principles in formation and dynamics of interacting electron transport chains. Although it was shown before that heterologous expression of MtrB in E. coli is possible, the results obtained in this study raise questions in terms of functionality of the expressed protein. Many studies showing proper integration of the Mtr pathway into E. coli but reveal by far lower reduction rates compared to S. oneidensis. The inaccurate folding discovered in this study using a series of Western blots provides an explanation for this behavior and motivates for further research in the particular field of β-barrel maturation. The experiments conducted to describe the dynamics of the electron transport routes leading to NO3- and NO2-reduction show an influence of the combination of the proteins NrfA and STC together with NO3-even in in vitro assays. Using cell suspension experiments it could be shown in vivo that the dynamic of the separation of NO3- and NO2-reduction is immediate. Since it was not possible to isolate and purify all reaction partners (NapA and NapB were unsuccessful) further efforts will be made to gain a complete description of this process.
