The main goals of the Central Project are the support of SPP projects on the one hand by measuring techniques for the characterization of mechanical properties of proteins, on the other hand to develop production and purification protocols for different ß-lactoglobuline bovine (BlgB) variants via systematic scouting investigations in the μL-scale. After scaling up of the production and purification procedure BlgB-variants are distributed to other SPP projects for comparative studies. Thus, the Central Project promotes interdisciplinary scientific exchange in the SPP consortium.The most frequently used model proteins in the SPP represent BlgB or its defined variants. Accordingly, these proteins will be produced and characterized in the second phase of the SPP in order to make them available for the working groups in the required form (in dissolved or crystal form). In this way, proteins that differ in a defined manner are available for comparative studies and enable a comprehensive scientific exchange.Therefore, the techniques and materials used are focused on the determination of mechanical properties of proteins with AFM and nanoindenter as well as the preparation and purification of different BlgB variants. Also alcohol dehydrogenase (ADH) and other techniques will be used in the Central Project.
