VIPP1 and VIPP2 are key players in the chloroplast unfolded membrane protein response (cpUMPR). Using Tur-boID, we identified interaction partners of VIPP1/2 that are upregulated under chloroplast stress (VPL1-11). Their association with VIPP1/2 will be verified by co-localization and reciprocal TurboID labelling. This will generate an interaction map to identify those VPLs that cluster around the function of VIPP1/2 in the cpUMPR. Subsequently, the interactions of these VPLs with VIPP1/2 in vitro and their functions in the cpUMPR in vivo will be analyzed. Stromal CDJ2 recruits its partner HSP70B in vitro to regulate the oligomeric state of VIPP1. Using a cdj2 knockout mutant, the importance of the dynamics of VIPP1 oligomer formation in vivo will be analyzed.
