Mitochondrial structure and dynamics in light and electron microscopy (Z03)

The CECAD Imaging Facility will support the CRC1218 with a Z-project (Z03). Despite our general support in established light and electron microscopy methods, we will set a focus on mitochondrial cristae structures in STED microscopy using SNAP constructs labelled with SNAP-Cell SiR. For electron microscopy, we will set up ApexII and HRP protocols for different mouse tissues, optimize HPF and freeze substitution for different tissues and C. elegans and move from TEM to serial section EM and serial section electron tomography to widen the spectrum to volume imaging. CLEM will be performed combining STED and TEM.
