The regulation of gene expression in the development of heart failure is not completely understood, and recent studies suggest that RNA methylation may play a significant role. Based on results from the previous funding period, we plan to further elucidate the mechanisms of m6A RNA methylation on gene regulation and on microRNA function. In addition, since H3K4 mono- and trimethylation have been proposed to modify gene/protein regulation, D04 aims to characterize the role of H3K4 methylation in the healthy and diseased heart and examine the effect of H3K4 methyltransferases and demethylases in cardiomyocyte-specific knockout mice.
